At the beginning, it is probably safe to uncheck all but the "Bayesian Test Mode". Once you find your mails to be no longer incorrectly categorized (IE ASSP has been trained with enough spam and non spam), you can look to turn off the Test Mode.

"A" records are better because CNAME records can cause an extra DNS lookup, which will slightly delay visitors to your website, and use extra bandwidth.

The "Add a sub domain" looks to function the same as "Add a C-Name (alias)".

ASSP also uses DNS Blacklists. If there is a RBL you want to use with ASSP, there is a field in ASSP web configurator that allows you to add more. There is no reason to use the built-in hMailserver blacklists.

Are you using a POP server address (for 3rd party email hosted) based on your domain names or one that this 3rd party provides? If it is one that this 3rd party provides then setting up external accounts for hMailserver will work fine without needing to set it to their mail server IP addresses.

I would suggest you use a dummy domain (EG example.com) for users of Destination.com to send out emails on and not have "Destination.com" domain set up on hmailserver. You can set in your email client that the FROM address to be whatever@Destination.com and that should work fine.
